According to a recent study conducted by the Pittsburgh
School of Medicine, rats with Impotence that were treated
with a gene therapy containing either of two nerve growth
factors were able to regain normal erectile function after
one month. These incredible findings are being presented
at the 10th annual meeting of the American Society of
Gene Therapy at the Washington State Convention and
Trade Center in Seattle. This extensive study was
conducted by Dr. Joseph Glorioso who is the chair of the
department of biochemistry and molecular genetics, and
Dr. Joel Nelson who is the chair of the department of
urology at the University of Pittsburgh School of Medicine.
These two researchers inserted either the gene for the glial
cell line derived neurotrophic factor also named GDNF or
the family ligand (neurturin) into a genetically engineered
herpes simplex virus. They then injected either of the
recombinant viruses into the damaged cavernous nerve of
rats. The important nerve growth promoter named GDNF
has been shown in many other studies to contribute to the
regeneration and survival of penile nerves. Neurturin is
also a nerve growth factor closely related to GDNF. The
placebo or the control mice experiment did receive the
virus but not the GDNF or neurterin genes inserted. After
one month of Impotence treatment, rats administered HSV
or GDNF exhibited significant recovery of intracavernous
pressure and systemic arterial pressure as opossed to
rats treated with the control virus with Impotence. Rats that
were treated with HSV-neurturin also exhibited significant
recovery compared with the control or untreated mice after
one month of Impotence treatment. The studies of
fluorescent proteins also showed that the delivered genes
had been effectively incorporated into the target nerve cells
which improved erectile function. According to Dr. Joseph
Glorioso, HSV delivery of GDNF and/or neurturin presents
a potentially important new approach for the treatment of
Impotence.
